How can I calculate fuel savings to determine if I can save money by slowing down when I commute to work?

Is there a math formula to use to calculate how much money can be saved on gas by slowing down?

I live almost 40 miles by the shortest route, but that means around 20 stoplights and taking back country roads. The highway route adds 15 miles and takes longer. Moving is not an option, nor is looking for a job closer to home. I have a fuel efficient vehicle, but am wondering what is the ideal speed to drive to save money, but won’t take me over an hour to get home from work after working 12-hour days. Also ride sharing is not an option either, as no one lives nearby and works the same hours.
Thanks to all who are writing. These all are great suggestions and will make it very hard to select a best answer.

Fill your tank. Reset your trip odometer. Drive normally. Refill your tank noting the gallons required. Divide miles travelled by gallons used. (This is MPG) Then repeat the process, but drive slower.

Most cars get the best mileage at 55mph although they used to do better at 60. next time you fill up write down your mileage. drive at least a quarter tank worth, then fill up and see how many gallons it took to fill up again. Also the fewer stops and starts you make gets better mileage.

in most car a fuel efficiat speed is around 55 mph or around 2200 rpm. it all depends on your hp and tranny match. the best thing u can do is figure out your mpg {fill up your tank till the pump nozzle clicks off,zero out your trip odometer or wright down the miles. drive it the normal route u take till half a tank is left,fill it up till nozzle clicks. wright down how many GALLONS not dollars u put in. then do the math. miles divided by gallons = mpg. now do the same thing at a slower speed till half a tank and repeat

Drive one route a specific number of trips, fill up, note the
number of gallons. Drive the other route the same number
of times, fill up, note the number of gallons. The lowest
number wins. For a fair test you must start each test with
the same amount of gas (as close as you can estimate it).
